PostDoc position in the department of Department of Biological Chemistry at Weizmann institute of Science in the group of Sarel Fleishman for 3 years. We are a new lab working on de novo design of protein function and are seeking highly motivated and capable postdoctoral fellows, with experimental or computational background (or both). Our research is aimed at understanding and mastering the design principles of protein functional sites. We test and enhance our capabilities by designing proteins that do not exist in nature to carry out desired functions. Our vision is to understand the principles of natural biomolecular function to the point where we can design any desired function from scratch. We currently focus on three research areas: 1. design of antibody binders from first principles. We have developed novel computational strategies to design antibodies to bind, in principle, any desired protein surface. We are continuing to improve the design strategy, and are now testing these antibodies experimentally. 2. design of specificity and multispecificity in the membrane. We are developing a unique platform based on in vitro evolution, deep mutational sequencing, and computational modeling to increase our understanding and ability to design specific and multispecific interactions in the membrane. 3. design of the first synthetic interaction network. We have designed several proteins to bind a target protein in a multispecifc manner, generating the first protein interaction network that is fully synthetic and thus unconstrained by evolution. We are now using this system to study how affinity and specificity emerge in small protein interaction networks. In all of these we use and develop new experimental and computational methodologies. The computational work uses the state-of-the-art Rosetta software for macromolecular modeling. Experimental strategies include: protein production and purification, cloning, deep mutational scanning, in vitro evolution using yeast cell-surface display, and binding experiments.
