Protein ubiquitylation is reversed by deubiquitylating enzymes (DUBs), which are specialized proteases that recognize the linkage between ubiquitin molecules in chains or between ubiquitin and protein substrates. Humans encode ~100 DUBs and, while it is appreciated that DUBs are important regulators of the ubiquitin system, the precise cellular roles of most DUBs are unknown.
