PROGRESSION OF HER-2 BREAST CARCINOMA: PATHWAYS AND THERAPY IN NEW IN VIVO MOUSE MODELS

This project is focused on the progression of HER2+ Breast Cancer (BC) and will study two mechanisms likely involved in multiorgan metastasis and therapy resistance: 1) the coexpression of full-length HER2 with variants, such as HER2Delta16 isoform (D16), associated with aggressive cancers; 2) the onset of HER2-loss variants.
Two original complementary mouse models will be used: a) an immunocompetent transgenic mouse model of mammary carcinogenesis co-expressing human HER2 and D16; b) a multiorgan metastasis model of HER2+ BC xenograft cells and clinical samples.
Activated pathways will be analyzed in untreated and anti-HER2 treated mice.
Search for new pathways and targets will be based on gene expression analysis of mammary glands during tumor onset, of tumor and metastases at different sites, and of mammospheres enriched for Cancer Initiating Cells (CIC).
Vaccines, antibodies and drug therapies, and combinations, targeting these pathways, will be studied.
