Study of the role of Polycomb Group Complex in determining cleft lip and palate

BACKGROUND. Cleft lip with or without cleft palate (CL/P) is one of the most common congenital malformations, caused by both genetic and environmental influences. In the last decade genetic studies identified a great number of candidate genes and loci contributing to clefting susceptibility. Our group showed allelic association between cleft lip and JARID2, a gene highly and specifically expressed at epithelial cells level of the laminae palatine. Recently, it has been reported that JARID2 interacts with the Polycomb-Repressive Complex 2 (PRC2) in regulating the expression patterns of developmental genes through modification of chromatin state.
AIMS. The goal of this project is to identify the genetic causes of orofacial clefts through a candidate gene association study. Particularly, we will investigate polymorphic variants of genes encoding proteins belonging to or interacting with PRC2 complex.
