Effects of lactate dehydrogenase inhibition on the pluripotency signatures of cancer cells

Cancer cells usually display increased glucose uptake and upregulated glycolytic pathway, which produces increased pyruvate and NADH levels. Excess NADH is re-oxidized by lactate dehydrogenase A (LDHA), a constantly up–regulated enzyme in cancer cells. LDHA is a target of HIF-1, which is induced by a hypoxic microenvironment, often found in solid tumor lesions. HIF-1 was found to also increase the expression of genes inducing the pluripotent phenotype. The cells exhibiting this phenotype are a major cause of anticancer treatment failure. They are often located in tumor regions characterized by hypoxia, where their survival is highly dependent on HIF-1 and LDHA. As a consequence, it can be hypothesized that LDHA inhibition in tumors could also cause the loss of the stem-like properties. The objective of this project is to verify this hypothesis by studying the effects of LDH small molecule inhibitors on the expression of genes responsible of the stemness properties of cancer cells.
