This project aims at the application of the dual laser optical tweezers to mechanical studies on single molecules and supramolecular systems involving the contractile and the cytoskeleton proteins of muscle and the membrane proteins responsible for the mechanosensitivity of neurons. A first objective of the research is the development and the characterization of a synthetic biomachine made of a single actin filament brought to interact with an array of myosin motors disposed on a nano-structured inorganic substrate. A second objective is the application of DLOT to the characterization of nociceptor neurons that are constitutively sensitive to mechanical stimulation. In collaboration with the Loboratory of Pharmacology of the Dipartimento Area Critica Medico-Chirurgica we will use neurons in culture to record the mechanical parameters of mechanosensing and its modulation by target molecules
