GDNF and SST are two neuromodulators expressed by nociceptive peptidergic primary afferent fibers of the spinal cord, implicated in the modulation of nociceptive transmission acting through specific receptors. The project will investigate the modulatory effects of GDNF/SST on sensory neurotransmission in two models of chronic pain: a zymosan-induced inflammatory pain model and a streptozotocin-induced type 1 diabetic (T1D) neuropathic pain model. The following aims will be addressed: AIM 1) To study the interactions between GFRa1/RET and SSTR2 in the same nociceptors and understand if the activation of one interfere on the activity of the other. AIM 2) To investigate the role of GDNF/SST in a zymosan-induced inflammatory pain model. AIM 3) To investigate the role of GDNF/SST in a streptozotocin-induced T1D pain model. A multidisciplinary experimental approach will be used: Ca2+ imaging, electrophysiology, light and electron microscopic immunohistochemistry, behavioural tests.
