Genetic engineering and development of oncolytic herpes simplex viruses retargeted to cancer-specific receptors

The aim of the project is the genetic engineering of oncolytic herpes simplex viruses (HSV), and their preclinical evalutation. HSV is a good oncolytic virus because it is a mild pathogen, it can be controlled with Acyclovir in a worst-case scenario, it kills cells. Sophisticated genetic engineering techniques are available. While the size of the genome (150Kbp) makes the genetic engineering rather complex, it actually represents an advantage, in that space is available for insertion of heterologous genes that enable more effective tumor oncolysis. Our laboratory has developed the technologies to modify HSV tropism and generate oncolytic-HSVs (o-HSVs) retargeted to cancer-specific receptors, and detargeted from normal receptors. The strategy consists in the modification of the receptor-binding glycoprotein gD through insertion of ligands to the cancer receptor, in our case the HER-2 receptor, overexpressed in about 30% of human mammary and ovary tumors.
