The main goals of this project are:
1)Evaluating the effectiveness of early and prolonged mu4D5 (the murine form of
trastuzumab)treatment in Δ16HER2-LUC mouse.
2)Establishing Δ16HER2-expressing cell lines from mammary tumors explanted from
Δ16HER2-LUC mice. These cell lines will represent a useful tool for studing the
oncogenic Δ16HER2 signaling pathways and the molecular events that contribute tracellular and transmembrane domains of Δ 16HER2(Δ16HuHuT) human splice variant, in comparison with HuHuT, encoding the fully
human wild-type extracellular and transmembrane domains of HER2.
4)Evaluating the response of Δ16HER2-LUC transgenic mouse to novel epitopebased
DNA vaccines, coding for chimeric proteins containing the Fc-region of mouse IgG2A and novel HER2 epitopes, identified using a phage-displayed collection human HER2 fragments.
