the development of a novel lipid-based sensors for dosing the activity of three specific enzymes that are biomarker of many solid tumours. Lipid vesicles (liposomes) will be designed and formulated in such a way that the interaction of the lipid embedded substrate with the target enzymes (5-fluorouracile, 5-FU) will induce lipid reorganization that will trigger an optical response. The experimental investigations will concern the synthesis of the lipids to be included in liposome formulations and the actual setup of sensors. Various mixed liposomes either using the various types of fluorescent lipids or exploiting non-labelled lipids and including a proper fluorescent probe in the formulation (at the lipid/water interface or in the internal aqueous phase) will be formulated. Lipid composition, fluorescent probe concentration, liposome concentration, kind of buffer and overall concentration conditions will be investigated for observing a proper and highly sensitive optical response.
