Immunoconjugates containing polynucleotide:adenosine glycosylases for the experimental therapy of B-lymphomas: study of the pathogenetic mechanisms of cancer cell intoxication and evaluation of antitumor efficacy

The research activity will be focused on the design and testing of immunotoxins (conjugates antibody/toxin) for the experimental therapy of B lymphomas. The plant toxins PNAG (polynucleotide:adenosine glycosilases) will be used as toxic compound.
Stages of research: 1- Design and construction of new immunotoxins specific for CD20 and CD22, obtained through chemical modification and conjugation of the two components, mAb and toxin, in order to obtain specific drugs for target lymphoma cells. The selected immunotoxins will be evaluated, in vitro, on target cell lines and on primary cultures of malignant lymphoma B cells. The immunotoxin antitumor activity will be also tested, in vivo, on a SCID mouse/B lymphoma cells model. 2- Study of the mechanisms of PNAG cytotoxicity on cancer cells: identification of the primary targets of PNAG in the cell and evaluation of the cell death pathways triggered.
