The present project will investigate the role of BARs in regulating CSE and CBS, the two main H2S-generating enzymes, in entero-hepatic tissues and in the cardiovascular system by molecular and pharmacological approaches. Mice deficient for FXR and TGR5 will be characterized for the expression of CSE, CBS and eNOS in the liver, heart, aorta and gastrointestinal tract. Human samples obtained from patients with liver and gastrointestinal disorders will be investigated for CBS and CSE.
Dissecting molecular pathways involved in regulation of CSE and CBS by BARs will be a major step in deciphering the physiology and pathophysiology of H2S and will allow the discovery of novel molecules of potential clinical application in the treatment of liver, gastrointestinal and cardiovascular disorders, including NASH and atherosclerosis, when a modulation of H2S generation could be of therapeutic utility.
