The enzymes of the metabolic pathway of thioredoxin play a fundamental role in the physiology of all organisms, and are involved in the pathology of several diseases. Thioredoxin Reductase is essential for the survival and the virulence of the human parasite P. falciparum. The enzyme has been cloned and heterologously expressed in E.coli, and our research project includes its structural and functional characterization, with special reference to the effect of known or putative inhibitors. These will be characterized by steady state and rapid kinetic methods (if applicable) and crystallization of the enzyme inhibitor complex will be attempted with the aim of obtaining their 3D structure.
