The Ca2+-mediated functional crosstalk between ER and mitochondria regulates the apoptotic response to several stimuli. Moreover, Ca2+ transfer to mitochondria and apoptosis is also influenced by the ER-mitochondria distance and tethering. Aim of this project is to evaluate the importance of the ER-mitochondria crosstalk in the modulation of apoptosis in different prostate cancer cell lines. Morphological and functional integrity, as well as the ER-mitochondria tethering, will be evaluated by confocal analysis and imaging, and subfractionation protocols. Cell sensitivity of different prostate cancer cell lines to several pro-apoptotic stimuli will be also evaluated by measuring Ca2+-exchange between ER and mitochondria.
