The overall objective of this project is to demonstrate the ability of isolated domains from mitochondrial (mt) aminoacyl-tRNA synthetases to rescue the mitochondrial defect caused by point mutations in mt tRNA genes in human cells. In particular, we will evaluate the effect of the isolated carboxy-terminal domain of human mt leucyl-tRNA synthetase and small fragments derived from it, in cybrid cells with the m.3243A>G mutation in tRNALeu(UUR), causing the severe Mitochondrial Encephalomyopathy with Lactic Acidosis and Stroke-like episodes (MELAS) syndrome.
