Study T cell subsets by flow polychromatic cytometry in patients with multiple sclerosis

CD8 MAIT cells play an important role in the pathogenesis of MS. Recent advances in flow cytometry, paralleled by the development of new software which enables the analaysis of complex data sets, enable the detailed study of the pehenotype and function of this cellular subset (activation status, adhesion molecule expression, cytokine and chemokine production, proliferation). In this project we will:

1)Use 9 colour flow cytometry for the accurate definition of these cells, both ex vivo and in vitro. Cells will also be sorted on a MoFlo for in vitro functional studies (see below). Data will be analyzed with FlowJo and InfiniCyt.

2)Perform the functional characterization of CD8+ MAIT T cell lines (proliferation following antigenic challenge, cytokine production, and transcriptional profiling).
