Biomedical devices have been introduced in several medical fields. Aim of the proposal is to investigate, in bone metastatic tumor treatment, the efficacy of nanocomposite hybrid hydrogels loaded with antineoplastic drugs. The proposal will be organized as following:
1) analysis of the biocompatibility of the nanocomposite hybrid hydrogels. Effects on cell toxicity, growth and differentiation will be carried out at short and long experimental times;
2) preclinical investigation of the in vitro and in vivo efficacy of the devices as a delivery systems for targeted release of antineoplastic/antiangiogenic drugs.
The cytotoxic/molecular effects of both the drugs and the hydrogel degradation products will be analyzed using human prostate and breast cancer cell lines, examples of bone metastasis, and human endothelial cells. The preclinical therapeutical efficacy of drugs released from the devices will be tested in vivo in animal models of subcutaneous-engrafted tumors in mice
