Role of PARP-1 on dendritic cell differentiation as possible target for immunological dysregulation-associated diseases

Dendritic cells are antigen presenting cells involved in innate and adaptative immunity. There is a great interest on their pathogenetic role on inflammatory and immunological-related diseases, such as multiple sclerosis (MS) as well as atherosclerosis. It has been demonstrated that DC are involved in epitope spreading, necessary event during progression of MS; moreover it is known that DC number increases in atherosclerotic lesions and in the symptomatic lesions. Poly [ADP-ribose] polymerase 1 (PARP-1) is a key nuclear enzyme in the regulation of gene expression which is expressed in DC during their differentiation. In Apo-/- mice model of atherosclerosis PARP-1 is associated to inflammatory responses. The aim of this project is to study in vitro pharmacological PARP-1 modulation on DC differentiation, phenotype and function
